Summary

FHIT gene abnormalities in hepatocellular carcinoma (HCC) were investigated. Aberrant FHIT transcripts were frequent in liver tissues but not causally linked to cancer development.

Background:

  • Alterations in the FHIT gene have been implicated in various cancers.

Purpose of the Study:

  • To investigate FHIT gene abnormalities in human hepatocellular carcinoma (HCC).
  • To determine if FHIT gene alterations are associated with HCC development.

Main Methods:

  • Analysis of eight liver cancer cell lines and 21 liver tissue samples (18 paired tumorous/non-tumorous).
  • Microsatellite polymorphism analysis, reverse transcription of FHIT mRNA, PCR amplification, and sequencing.
  • Detection of loss of heterozygosity and FHIT transcript variants.

Main Results:

  • No loss of heterozygosity at chromosome 3p14.2 was detected.
  • Normal FHIT transcripts were expressed in most samples.
  • Aberrant FHIT transcripts, likely alternatively spliced, were found in a high frequency of tumorous and non-tumorous tissues.

Conclusions:

  • FHIT gene transcript abnormalities are common in liver tissues, including HCC.
  • These transcript abnormalities may not be directly causally related to hepatocarcinogenesis.
