Summary

Nicotine, a major component of cigarette smoke, does not appear to negatively impact thyroid function or hormone levels in rats. This study suggests nicotine is not the cause of thyroid issues observed in human smokers.

Background:

  • Smoking is linked to more severe hypothyroidism and altered thyroid markers in women.
  • Thiocyanate from smoke is a known goitrogen, but doesn't explain all observed thyroid effects.
  • The role of nicotine, another major cigarette component, on thyroid function in vivo is unknown.

Purpose of the Study:

  • To investigate the effects of nicotine on thyroid function and thyroid hormone action in vivo.
  • To determine if nicotine contributes to the adverse thyroid effects associated with smoking.

Main Methods:

  • Rats (euthyroid, subclinically hypothyroid, and L-thyroxine treated) received nicotine infusions for 7 days.
  • Assessed serum thyroid hormones (T4, T3), TSH, cholesterol, and hepatic malic enzyme activity.

  • Measured outer-ring 5'deiodinase (5'D-I) activity in liver and kidneys, and intrathyroidal iodine metabolism.
  • Main Results:

    • Nicotine infusion did not alter serum T4, T3, TSH, or cholesterol levels in any rat group.
    • Nicotine did not affect liver or kidney 5'D-I activity or hepatic malic enzyme activity.
    • No significant impact of nicotine on thyroid function markers or thyroid hormone action was observed.

    Conclusions:

    • Nicotine administration does not adversely affect thyroid function or thyroid hormone action in rats.
    • These findings strongly suggest nicotine is not responsible for the thyroid dysfunction observed in human smokers.
