Does the T-locus in the mouse include ribosomal DNA?

K C Atwood, S Gluecksohn-Waelsch, M T Yu

    Summary

Ribosomal DNA (rDNA) does not influence the T-locus genotype in mice. Researchers found no link between rDNA and chromosome 17, which carries the T-locus, simplifying future genetic studies.

    Area of Science:

    • Genetics
    • Molecular Biology
    • Genomics

    Background:

    • The T-locus in mice is known for unique allelic properties.
    • Ribosomal DNA (rDNA) has been considered a potential factor influencing genetic loci.
    • Understanding the relationship between rDNA and specific genetic loci is crucial for genetic research.

    Purpose of the Study:

    • To investigate the relationship between the proportion of ribosomal DNA (rDNA) and the T-locus genotype in the mouse genome.
    • To determine if rDNA is located on chromosome 17, which harbors the T-locus.

    Main Methods:

    • Analysis of the proportion of rDNA in the mouse genome.
    • Hybridization in situ using 125I-RNA to detect rDNA presence on specific chromosomes.
    • Genotyping of the T-locus in relation to rDNA proportion.

    Main Results:

    • No systematic relationship was found between the proportion of rDNA and the T-locus genotype.
    • Hybridization in situ experiments did not label chromosome 17 (T-locus carrier) with rDNA.
    • The average number of genes for rRNA precursor was 153 per haploid genome, with significant individual variation.

    Conclusions:

    • rDNA can be excluded as a factor explaining the unique properties of T-locus alleles.
    • Future research on the T-locus does not need to consider rDNA.
    • The significant individual variation in rRNA precursor gene number warrants further investigation.
