Background:

  • Mast cells play roles in immune responses, tumor progression, and vascularization.
  • Investigating mast cells in situ within routinely processed tissues is challenging due to detection difficulties.

Observation:

  • A morphometric point-counting technique was used to quantify mast cell tissue density in Giemsa-stained skin biopsy sections.
  • This method proved accurate and reproducible for mast cell density determination.

Findings:

  • Mast cell density was significantly greater in invasive melanoma (mean 0.61 vol.%) compared to benign melanocytic nevi (mean 0.33 vol.%) and melanoma in situ (mean 0.5 vol.%).
  • Six of 15 invasive melanoma cases exhibited mast cell densities exceeding 0.6 vol.%, a threshold not surpassed in nevi or melanoma in situ (p < 0.02).
