    Summary

Optimal cryopreservation of human lymphocytes requires careful condition selection. Freezing cells in autologous serum with 12.5% dimethylsulfoxide (DMSO) yielded the best results for maintaining cell viability after thawing.

    Area of Science:

    • Immunology
    • Cell Biology
    • Cryobiology

    Context:

    • Lymphocytes are crucial for immune responses.
    • Cryopreservation is essential for long-term storage of lymphocytes.
    • Maintaining high cell viability post-thaw is critical for lymphocytotoxic assays.

    Purpose:

    • To determine optimal conditions for freezing human lymphocytes.
    • To evaluate the impact of cryoprotectants, media composition, and other parameters on lymphocyte viability.
    • To minimize cell death during the freeze-thaw process.

    Summary:

    • The study investigated various factors influencing the cryopreservation of human lymphocytes.
    • Dimethylsulfoxide (DMSO) concentration, serum type, and medium composition were key parameters.
    • The most favorable results were achieved using 12.5% DMSO in autologous serum, with good outcomes also observed in AB serum and 20% bovine serum in Eagle's medium.

    Impact:

    • Establishes optimal cryopreservation protocols for human lymphocytes.
    • Enhances the reliability of immunological assays relying on viable lymphocytes.
    • Provides valuable data for biobanking and clinical applications involving frozen lymphocytes.
