Area of Science:

  • Biochemistry
  • Pathophysiology
  • Free radical chemistry

Background:

  • Peroxynitrite is a toxic metabolite formed from nitric oxide and superoxide.
  • It oxidizes biomolecules and modifies tyrosine residues, creating detectable nitrotyrosines.
  • Free radicals, including peroxynitrite, are implicated in various pathological states.

Purpose of the Study:

  • To highlight the role of peroxynitrite in disease pathophysiology.
  • To discuss the oxidative and nitrating capabilities of peroxynitrite.
  • To explore the potential of peroxynitrite scavengers as therapeutic agents.

Main Methods:

  • Review of existing literature on peroxynitrite.
  • Analysis of peroxynitrite's chemical reactivity with biomolecules.
  • Investigation of peroxynitrite's involvement in disease mechanisms.

Main Results:

  • Peroxynitrite oxidizes proteins, lipids, carbohydrates, and nucleic acids.
  • It causes hydroxylation and nitration, forming nitrotyrosines.
  • Peroxynitrite is linked to inflammation, atherosclerosis, and ischemia-reperfusion injury.

Conclusions:

  • Peroxynitrite is a significant contributor to various disease pathologies.
  • Nitrotyrosine formation serves as an in vivo marker for peroxynitrite activity.
  • Peroxynitrite scavengers represent a promising therapeutic avenue for related diseases.
