Summary

Background:

  • Sweat gland carcinomas are rare skin tumors with unknown causes and molecular underpinnings.
  • Understanding the molecular basis of these tumors is crucial for diagnosis and treatment.

Purpose of the Study:

  • To investigate the presence and types of p53 mutations in various histologic types of sweat gland carcinomas.
  • To explore the potential role of UV radiation and p53 mutations in the malignant progression of these tumors.

Main Methods:

  • Analysis of p53 mutations in 16 sweat gland carcinomas using single-stranded conformation polymorphism (SSCP) and direct DNA sequencing.
  • Histologic typing of all analyzed tumors.

Main Results:

  • p53 mutations were detected in 5 out of 16 (31%) sweat gland carcinomas.
  • Four mutations were G:C-->A:T transitions, and one was a deletion.
  • Three G:C-->A:T mutations occurred at dipyrimidine sequences, suggesting a potential role for UV light.
  • p53 mutations were found in carcinoma but not adenoma portions of two spiradenocarcinomas, indicating association with malignant progression.

Conclusions:

  • p53 mutations are present in a subset of sweat gland carcinomas.
  • UV radiation may be a contributing factor in the etiology of some sweat gland carcinomas.
  • p53 mutations appear to be involved in the malignant progression of these rare skin tumors.
