Summary
CRCS and mineral trioxide aggregate (MTA) demonstrated the lowest cytotoxicity among root canal sealers and root-end filling materials, respectively. Gallium GF2 also showed favorable results as a retrofilling material.
Area of Science:
- Biomaterials Science
- Dental Materials
- Cell Biology
Background:
- Root canal sealers and root-end filling materials are crucial in endodontic treatments.
- Assessing the cytotoxicity of these materials is essential for patient safety and treatment success.
- Previous evaluations have varied, necessitating further comparative studies.
Purpose of the Study:
- To evaluate and compare the in vitro cytotoxicity of commonly used root canal sealers and root-end filling materials.
- To identify the most biocompatible materials for endodontic applications.
Main Methods:
- An in vitro cell culture model using human gingival fibroblasts and L-929 cells was employed.
- Cytotoxicity was assessed using the MTT assay (mitochondrial enzyme activity) and the CV assay (cell numbers).
- Materials tested included root canal sealers (Endomet, CRCS, AH26) and root-end filling materials (Amalgam, Gallium GF2, Ketac Silver, MTA, Super-EBA), plus All-Bond-2.
Main Results:
- CRCS exhibited the least cytotoxicity among root canal sealers, followed by Endomet and AH26.
- Mineral trioxide aggregate (MTA) showed no cytotoxicity; Gallium GF2 displayed minimal cytotoxicity as a root-end filling material.
- Ketac Silver, Super-EBA, and Amalgam demonstrated higher cytotoxicity, as did All-Bond-2.
Conclusions:
- CRCS is recommended as the optimal root canal sealer due to its low cytotoxicity.
- MTA is the preferred root-end filling material based on its biocompatibility.
- Gallium GF2 presents a favorable alternative for retrofilling applications.
