Area of Science:

  • Medicinal Chemistry
  • Pharmacology

Background:

  • 1,4-dihydropyridines are a known class of calcium channel blockers.
  • Nifedipine is a widely used 1,4-dihydropyridine derivative.

Purpose of the Study:

  • To synthesize novel 1,4-dihydropyridine derivatives incorporating a coumarin moiety.
  • To evaluate the inotropic, chronotropic, and calcium antagonist activities of these new compounds.

Main Methods:

  • Synthesis of 1,4-dihydropyridine compounds with coumarin at the 4-position.
  • Pharmacological evaluation of inotropic, chronotropic, and calcium antagonist effects.

Main Results:

  • The synthesized compounds exhibited decreased activity on myocardial and vascular parameters compared to nifedipine.

Related Experiment Videos

  • Selective inhibition of cardiac contractility and frequency was observed.
  • Conclusions:

    • Replacing the o-nitrophenyl group of nifedipine with coumarin or phenylcoumarin systems reduces overall activity.
    • The synthesized coumarin-bearing 1,4-dihydropyridines demonstrate selective cardiac effects.
