Summary

New simple sequence repeat (SSR) markers were developed for potato (Solanum tuberosum L.) genetic studies. These SSR markers offer high polymorphism and are recommended to replace Restriction Fragment Length Polymorphisms (RFLP) for future research.

Purpose of the Study:

  • To develop and characterize novel SSR markers for Solanum tuberosum L.
  • To assess the polymorphism and utility of these SSR markers for genetic mapping in potato.

  • Extraction of Solanum tuberosum L. DNA sequences with SSR motifs from databases and libraries.
  • Enrichment of DNA libraries using triplex affinity capture or single-strand hybridization selection.
  • Design and synthesis of 112 primer pairs, followed by amplification and polymorphism analysis in potato clones.
