Background:

  • AF10, a human B cell line, is a mycoplasma-free variant derived from the U266 IgE myeloma cell line.
  • Interleukin-17 (IL-17) is a cytokine with known roles in inflammation and immunity.

Purpose of the Study:

  • To investigate the presence of human interleukin-17 (hIL-17) messenger RNA (mRNA) in the AF10 human B cell line.
  • To explore the potential functional activity of expressed hIL-17.

Main Methods:

  • Total RNA was isolated from AF10 cells.
  • Reverse transcription polymerase chain reaction (RT-PCR) was employed to detect hIL-17 mRNA.
  • Recombinant hIL-17 (rhIL-17) expressed in E. coli was used to stimulate mouse fibroblast 3T3 cells.
