    Background:

    • May-Hegglin anomaly is a rare genetic disorder affecting blood cell development.
    • Megakaryocytes are essential for platelet production, and their polyploidization is a key process in their maturation.

    Purpose of the Study:

    • To investigate the polyploidization process of megakaryocytes in patients with May-Hegglin anomaly.
    • To understand the potential contribution of megakaryocyte abnormalities to the pathological platelet sequestration observed in this condition.

    Main Methods:

    • Bone marrow aspirates from 3 patients with May-Hegglin anomaly were analyzed.
    • Combined application of cytophotometric DNA content determination and in vitro autoradiography with 3H-thymidine (3H-TdR) labeling was used.

    Main Results:

    • A significant increase in progenitor cell influx into the megakaryocytic system was observed.
    • A decrease in maturation capacity from type II to type III megakaryocytes was noted.
    • Despite these changes, the polyploidization activity, assessed by 3H-TdR labeling and DNA content, was normal.

    Conclusions:

    • The observed abnormalities in progenitor cell influx and megakaryocyte maturation may contribute to pathological platelet sequestration in May-Hegglin anomaly.
    • Megakaryocyte polyploidization itself appears to be unaffected in patients with May-Hegglin anomaly.
