Summary

This study details the synaptonemal complex (SC) formation during meiosis in Locusta migratoria spermatocytes. We observed asynchronous chromosome pairing and SC fragmentation, with shorter chromosomes completing synapsis first and maintaining morphology longer.

Area of Science:

  • Cell Biology
  • Genetics
  • Microscopy

Background:

  • Meiotic prophase is crucial for accurate chromosome segregation.
  • The synaptonemal complex (SC) facilitates homologous chromosome pairing and recombination.
  • Understanding SC dynamics is key to comprehending meiotic fidelity.

Purpose of the Study:

  • To elucidate the synaptic process during meiotic prophase in Locusta migratoria.
  • To characterize the assembly, progression, and breakdown of the synaptonemal complex (SC).
  • To analyze the role of chromosome length in synapsis and SC fragmentation.

Main Methods:

  • Analysis of synaptonemal complex (SC) spreads.
  • Light microscopy (LM) and electron microscopy (EM) techniques.
  • Observation of spermatocytes from Locusta migratoria.

Related Experiment Videos

Main Results:

  • Unpaired axial elements assemble during leptotene.
  • Synapsis initiates primarily at chromosome ends during zygotene, with shorter chromosomes synapsing faster.
  • Pachytene shows complete synapsis; SC fragmentation begins asynchronously during diplotene, affecting longer chromosomes first.

Conclusions:

  • Chromosome length influences the timing and progression of synapsis and SC fragmentation.
  • Asynchronous events in SC assembly and breakdown are characteristic of Locusta migratoria meiosis.
  • Detailed ultrastructural analysis provides insights into conserved meiotic mechanisms.
