    Background:

    • Droperidol is an antipsychotic medication with known cardiovascular effects.

    Purpose of the Study:

    • To investigate the mechanism of action underlying droperidol's negative chronotropic effect.
    • To determine if droperidol interacts with cholinergic, adrenergic, or calcium channel pathways.

    Main Methods:

    • Experiments were conducted on isolated rabbit atria and guinea-pig vas deferens.
    • Direct injections of droperidol into the sinus node artery of anesthetized dogs were performed.
    • Assays included assessment of cholinergic and adrenergic antagonism, calcium flux, and responses to isoproterenol and dopamine.

    Main Results:

    • Droperidol did not exhibit cholinergic action or beta-adrenergic antagonism.

    • Alpha-adrenergic antagonism may play a role, but the main effect on heart rate is likely due to sinus impulse exit blockade.
