Summary

Dialysis related amyloidosis (DRA) treatment may benefit from Lixelle hemoperfusion, which removes beta2-microglobulin (beta2M) and inflammatory cytokines. This adsorbent shows high efficiency in clearing key cytokines linked to DRA development.

Area of Science:

  • Nephrology
  • Biomedical Engineering
  • Immunology

Background:

  • Dialysis-related amyloidosis (DRA) is a long-term complication in hemodialysis patients.
  • Beta2-microglobulin (beta2M) levels do not directly correlate with DRA occurrence.
  • Inflammatory cytokines like IL-1, IL-6, and TNFalpha are implicated in DRA pathogenesis.

Purpose of the Study:

  • To investigate the in vitro adsorptive capacity of the Lixelle adsorbent for various inflammatory cytokines.
  • To evaluate the potential of Lixelle in removing both beta2M and inflammatory cytokines for DRA treatment.

Main Methods:

  • In vitro study using the Lixelle adsorbent.
  • Measurement of adsorptive rates for beta2-microglobulin (beta2M) and specific inflammatory cytokines (IL-1beta, IL-1RA, IL-6, IL-8, TNFalpha, sTNFr).

Related Experiment Videos

Main Results:

  • Lixelle demonstrated high adsorptive rates for IL-1beta (98.5%), IL-1RA (98.0%), IL-6 (82.9%), and IL-8 (99.9%).
  • Lower adsorptive rates were observed for TNFalpha (31.2%) and sTNFr (46.1%).
  • Adsorptive efficiency decreased with increasing molecular weight of the cytokines.

Conclusions:

  • The Lixelle adsorbent effectively removes beta2M and a range of inflammatory cytokines.
  • Simultaneous removal of beta2M and inflammatory cytokines by Lixelle may be a crucial therapeutic strategy for managing DRA.
