Flow cytometry offers a more comprehensive method for assessing cell proliferation using the Ki-67 antibody in solid tumors. This technique enhances prognostic insights beyond traditional microscopic observation.
Area of Science:
- Oncology
- Immunohistochemistry
- Cell Biology
Context:
- Monoclonal antibody Ki-67 is a key marker for cell proliferation.
- Current studies on solid tumors predominantly use microscopy (immunohistochemistry).
- Microscopic methods offer limited information for Ki-67 analysis in solid tumors.
Purpose:
- To introduce a DNA/Ki-67 double staining method for flow cytometry.
- To demonstrate the application of flow cytometry for Ki-67 analysis in solid tissues and cultured cells.
- To discuss the clinical significance and utility of flow cytometric Ki-67 studies.
Summary:
- A novel method for DNA/Ki-67 double staining in solid tissues and cultured cells for flow cytometry is presented.
- This approach overcomes limitations of traditional microscopic immunohistochemistry.
- The study validates flow cytometry as a powerful tool for Ki-67 assessment.
Impact:
- Enhances the evaluation of cell proliferative activity in neoplastic diseases.
- Provides more detailed prognostic information compared to conventional methods.
- Expands the clinical utility of Ki-67 as a biomarker.
Abstract:
Monoclonal antibody Ki-67 is a useful and easy means for evaluating cell proliferative activity. Furthermore, it appears to provide valuable prognostic informations in some neoplastic diseases. However, almost all studies on solid tumors were performed only by microscopic observation using immunohistochemistry, and not by flow cytometry. Therefore, the information deduced from the findings is limited. The method of DNA/Ki-67 double staining of solid tissue and cultured cells for flow cytometry is shown, and the significance and the usefulness of flow cytometric study of Ki-67 in clinical use are discussed.
