Insights

Cytokines like TNF and IL-1 boost cell surface Intercellular Adhesion Molecule 1 (ICAM-1) on melanoma cells. These cytokines also trigger soluble ICAM-1 release, potentially linking tumor immunity to elevated ICAM-1 in patients.

Area of Science:

  • Immunology
  • Cell Biology
  • Oncology

Background:

  • Intercellular Adhesion Molecule 1 (ICAM-1) plays a role in immune responses.
  • Elevated soluble ICAM-1 is observed in melanoma patients, but its source is unclear.
  • Melanoma cell lines offer a model to study ICAM-1 regulation.

Purpose of the Study:

  • To investigate how cytokines regulate ICAM-1 expression on human melanoma cells.
  • To determine if melanoma tumors release soluble ICAM-1 in vivo.
  • To explore the potential source of elevated soluble ICAM-1 in melanoma patients.

Main Methods:

  • Utilized the human melanoma cell line A375M.
  • Treated cells with cytokines including Interleukin 1 (IL-1), Interleukin 6 (IL-6), Tumor Necrosis Factor (TNF), and gamma-interferon (IFN-γ).
  • Measured cell surface and soluble ICAM-1 levels; analyzed serum from tumor-bearing mice.

Main Results:

  • Unstimulated A375M cells expressed cell surface ICAM-1 but minimal soluble ICAM-1.
  • IL-1, IL-6, TNF, and IFN-γ increased cell surface ICAM-1 expression.
  • TNF, IL-1, and IFN-γ induced the release of soluble ICAM-1.
  • Soluble human ICAM-1 was detected in serum from mice bearing human melanoma tumors, correlating positively with tumor weight.

Conclusions:

  • Cytokines significantly regulate both cell surface and soluble ICAM-1 expression in melanoma cells.
  • Melanoma tumors can be a source of soluble ICAM-1.
  • Cytokine-induced release of ICAM-1 from tumors may be relevant to immune recognition in melanoma.

Metastasis

Metastasis is the spread of cancer cells from the original site to distant locations in the body. Cancer cells can spread via blood vessels (hematogenous) as well as lymph vessels in the body.
Epithelial-to-Mesenchymal Transition
The epithelial-to-mesenchymal transition or EMT is a developmental process commonly observed in wound healing, embryogenesis, and cancer metastasis. EMT is induced by transforming growth factor-beta (TGF-β) or receptor tyrosine kinase (RTK) ligands, which further...

Cell Adhesion Molecules - Types and Functions

Cell adhesion molecules (CAMs) are pivotal to multicellularity and the coordinated functioning of tissues and organ systems. They enable physical interactions between cells and provide mechanical strength to tissues. They also function as receptors for signal transmission across the plasma membrane. The CAMs are broadly classified into four families - integrins, cadherins, selectins, and immunoglobulin-like CAMs (IgCAMs).
CAM Families
The Integrin family of proteins is primarily  involved in a...

Immunoglobulin-like Cell Adhesion Molecules

Immunoglobulin-like cell adhesion molecules or Ig-CAMs are a versatile group of cell surface glycoproteins belonging to the immunoglobulin protein superfamily. Ig-CAMs possess the characteristic immunoglobulin protein domains and other domains such as the fibronectin type III domain. The Ig domains are glycosylated to varying degrees in different Ig-CAMs.
Ig-CAMs exhibit either homophilic binding (to other Ig-CAMs) or heterophilic binding (to other ligands such as integrins). While most Ig-CAMs...
