Insights
A new liposome-based immunoassay (LIA) for measuring total complement activity (CH50) was evaluated. This assay offers reliable results for healthy adults, establishing new reference values.
Area of Science:
- Clinical Immunology
- Diagnostic Assay Development
Background:
- Total complement activity (CH50) is a critical indicator of the classical complement pathway.
- Accurate CH50 measurement is essential for diagnosing and monitoring various immune-related conditions.
- Existing methods for CH50 assessment may have limitations in automation and standardization.
Purpose of the Study:
- To evaluate the performance of a new, commercially available automated homogeneous liposome-based immunoassay (LIA) for measuring CH50.
- To establish reference values for CH50 in an adult population using the new LIA method.
- To assess the precision, accuracy, carry-over, and linearity of the CH50 LIA.
Main Methods:
- Serum samples from 100 healthy adult donors were analyzed using the CH50 WAKO LIA.
- The liposome-based immunoassay was assessed for within-run and between-run precision.
- Accuracy, carry-over, and linearity of the assay were also evaluated.
Main Results:
- The study successfully established reference values for CH50 in the adult population using the new LIA.
- The automated homogeneous liposome-based immunoassay demonstrated acceptable precision and accuracy.
- Linearity and carry-over were assessed, providing key performance data for the new assay.
Conclusions:
- The CH50 WAKO LIA is a viable automated method for measuring total complement activity.
- The established reference values provide a crucial benchmark for clinical interpretation in adult populations.
- This new assay facilitates standardized and efficient CH50 assessment in clinical immunology laboratories.
Abstract:
The Immunology department of Garcia de Orta Hospital, has evaluated a new automated homogeneous liposome-based immunoassay (L.I.A.), for measuring total complement activity, which has become recently commercially available from WAKO (Osaka, Japan), (CH50 WAKO). Serum samples from 100 healthy voluntary donors were assayed, with liposome-based assay. The reference values in our adult population concerning CH50 were established. Within-run and between-run precision, accuracy, carry-over and the linearity of the new method were also assessed.
