Insights
A novel serum-free medium effectively supports human lymphocyte cultivation, showing enhanced mitogen potency for scientific, diagnostic, and therapeutic applications.
Area of Science:
- Immunology
- Cell Biology
- Biotechnology
Background:
- Optimizing human peripheral blood lymphocyte (HPBL) cultivation is crucial for research and clinical applications.
- Traditional methods often rely on serum-containing media, which can introduce variability and safety concerns.
- Serum-free media offer a controlled environment for lymphocyte culture.
Purpose of the Study:
- To compare the efficacy of a serum-free medium (Hybris-2) against a standard serum-containing medium (RPMI 1640 with FBS) for HPBL cultivation.
- To determine optimal mitogen concentrations for lymphocyte proliferation in both media types.
- To evaluate the impact of serum-free conditions on lymphocyte subset dynamics and marker expression.
Main Methods:
- Human peripheral blood lymphocytes were cultured in Hybris-2 (serum-free) and RPMI 1640 (10% FBS).
- Cultures were stimulated with phytohemagglutinin (PHA) and interleukin-2 (IL-2).
- Optimal PHA concentrations were determined for each medium type.
- Lymphocyte proliferation, T cell subset (CD3+, CD4+) expansion, and marker dynamics were analyzed.
Main Results:
- Optimal phytohemagglutinin concentration was significantly lower in serum-free medium (0.5 µg/ml) compared to serum-containing medium (5 µg/ml).
- Both PHA and IL-2 demonstrated greater potency in stimulating lymphocyte proliferation in the serum-free Hybris-2 medium.
- Robust proliferation of CD3+ and CD4+ T lymphocytes was observed in both culture conditions.
- While marker dynamics were generally similar, significant inter-donor variability was noted.
Conclusions:
- The developed serum-free medium (Hybris-2) is a viable alternative for human lymphocyte cultivation.
- Serum-free conditions enhance mitogen efficacy, potentially reducing reagent costs and improving culture efficiency.
- This serum-free medium holds promise for scientific research, diagnostic assays, and therapeutic cell manufacturing.
Abstract:
We compared the cultivation of human peripheral blood lymphocytes in serum-free medium Hybris-2 and RPMI 1640 medium with 10% fetal bovine serum in the presence of phytohemagglutinin and interleukin-2. The optimal concentration of phytohemagglutinin significantly differed in serum-free and serum-containing media (0.5 and 5 microg/ml, [corrected] respectively). Both mitogens were more potent in stimulating the proliferation of lymphocytes in serum-free medium than in serum-containing medium. Strong proliferation of CD3(+) and CD4(+) T lymphocytes was observed in both media. The dynamics of other markers was similar in serum-free and serum-containing media. However, significant differences were revealed between individual donors. Our results indicate that the developed serum-free medium may be used in lymphocyte cultivation for scientific, diagnostic, and therapeutic purposes.
