Insights
Uncoupling protein 2 (UCP2) expression is decreased in women with unexplained recurrent spontaneous abortion (URSA). Lower UCP2 levels correlate with altered cytokine balance, suggesting a role in early pregnancy loss.
Area of Science:
- Immunology
- Reproductive Biology
- Cellular Metabolism
Background:
- Unexplained recurrent spontaneous abortion (URSA) poses a significant challenge in reproductive medicine.
- Macrophage function and cytokine profiles are implicated in early pregnancy loss.
- The role of uncoupling protein 2 (UCP2) in the context of URSA remains incompletely understood.
Purpose of the Study:
- To investigate the relationship between uncoupling protein 2 (UCP2) expression and the interleukin-10/interferon-gamma (IL10/IFNgamma) ratio in decidual macrophages of patients with URSA.
- To elucidate the potential role of UCP2 in regulating macrophage activity and cytokine secretion in URSA.
Main Methods:
- Decidual macrophages were isolated from women with URSA and controls using magnetic cell sorting (MACS).
- UCP2 expression was quantified via flow cytometry.
- Cytokine secretion (IL10 and IFNgamma) was assessed using enzyme-linked immunosorbent spot-forming (ELISPOT) assays.
Main Results:
- Women with URSA exhibited significantly lower UCP2 expression on decidual macrophages compared to controls (136±25 vs 201±31, P<0.01).
- UCP2 expression was positively correlated with the IL10/IFNgamma ratio in these macrophages (r=0.73, P<0.01).
Conclusions:
- Decreased UCP2 expression in decidual macrophages may be a characteristic feature of URSA.
- UCP2 appears to play a role in modulating macrophage cytokine secretion, potentially contributing to the pathophysiology of spontaneous abortion.
Objective:
To investigate the relationship between the expression of uncoupling protein 2 (UCP2) and ratio of interleukin-10/interferon-gamma (IL10/IFNgamma) in the macrophages from patients with unexplained recurrent spontaneous abortion (URSA).
Methods:
Twelve women undergoing selective termination of normal early pregnancy (control) and 11 having URSA were included in this study. Magnetic cell sorting (MACS) was used to isolate the macrophages in the decidua, and the expression of UCP2 was detected with flow cytometry. Cytokine (IL10 and IFNgamma) secretion by the macrophages was detected by enzyme-linked immunosorbent spot-forming (ELISPOT) cell assay.
Results:
Compared to the control group, the women with URSA showed significantly decreased expression of UCP2 on decidual macrophage (136-/+25 vs 201-/+31, P<0.01), and the expression of UCP2 was positively correlated to the ratio of IL10/IFNgamma(r=0.73, P<0.01).
Conclusion:
UCP2 may play an important role in the regulation of macrophage activity and cytokine secretion to contribute to spontaneous abortion.
