In 1928, a German botanist Emil Heitz observed the moss nuclei with a DNA binding dye. He observed that while some chromatin regions decondense and spread out in the interphase nucleus, others do not. He termed them euchromatin and heterochromatin, respectively. He proposed that the heterochromatin regions reflect a functionally inactive state of the genome. It was later confirmed that heterochromatin is transcriptionally repressed, and euchromatin is transcriptionally active chromatin.

Sus antecedentes:

  • Los genes Hox de los vertebrados son cruciales para establecer el patrón del cuerpo anterior-posterior.
  • La activación del gen Hox ocurre en una secuencia espaciotemporal que refleja su organización de clúster genómico (colinealidad).
  • Los mecanismos moleculares que gobiernan esta activación espacio-temporal precisa siguen siendo en gran medida desconocidos.

Objetivo del estudio:

  • Investigar los mecanismos reguladores subyacentes a la activación espacio-temporal de los genes Hox de los vertebrados.
  • Identificar los posibles elementos reguladores responsables de iniciar y mantener la colinealidad del gen Hox.

Principales métodos:

  • Caminar genómico combinado con sucesivas inserciones de transgenes aguas arriba del complejo HoxD.
  • Generación de una serie de eliminaciones dentro de la región aguas arriba identificada.
  • Análisis de los patrones de activación del gen Hox y las consecuencias fenotípicas (transformaciones homeóticas).

Principales resultados:

  • Una deficiencia significativa aguas arriba del complejo HoxD indujo transformaciones homeóticas posteriores.
  • Esta deficiencia también dio lugar a una activación más temprana de los genes Hoxd.
  • Estos hallazgos señalan un elemento regulador esencial para el patrón de activación temprana del gen Hox.

Conclusiones:

  • Un elemento regulador ubicado aguas arriba del complejo HoxD juega un papel crítico en el establecimiento de la activación temprana colineal espacio-temporal de los genes Hox.
  • Este elemento parece estar involucrado en iniciar una conformación represiva que se alivia secuencialmente.
  • Comprender este mecanismo proporciona información sobre los principios fundamentales del desarrollo de los vertebrados y la regulación génica.
