Sus antecedentes:

  • Los transductores de señales y los activadores de la transcripción (STAT) son factores de transcripción clave que median las respuestas celulares a las citoquinas y los factores de crecimiento.
  • La activación persistente de Stat3 se observa en numerosos cánceres humanos y líneas celulares transformadas, donde juega un papel en la transformación y la resistencia a la apoptosis.

Objetivo del estudio:

  • Para investigar el potencial oncogénico de una molécula Stat3 constitutivamente activa.
  • Para determinar si la activación de Stat3 por sí sola es suficiente para la transformación celular.

Principales métodos:

  • Se utilizó la mutagénesis dirigida al sitio para sustituir dos residuos de cisteína en el dominio SH2 de Stat3, creando una molécula Stat3-C constitutivamente activa.
  • Se evaluaron las propiedades funcionales de Stat3-C, incluida la dimerización, la unión al ADN y la actividad transcripcional.
  • La transformación celular se evaluó mediante ensayos de formación de colonias de agar blando y formación de tumores en ratones desnudos.
