Reprogramming alters the gene expression in somatic cells, transforming them into induced pluripotent stem (iPS) cells over several generations. The ability of induced pluripotent stem cells or iPSCs to differentiate into most body cell types has stimulated repair and regenerative medicine research over the past few decades. iPSC-derived blood cells, hepatocytes, beta islet cells, cardiomyocytes, neurons, and other cell types can repair injuries or regenerate damaged tissue in diseases such as diabetes and neurodegenerative disorders.

Sus antecedentes:

  • La diferenciación celular restringe progresivamente el potencial de desarrollo durante el desarrollo animal.
  • En el sistema nervioso central (SNC), las células madre multipotentes generan precursores que finalmente se diferencian en neuronas o células gliales.

Objetivo del estudio:

  • Investigar el potencial de las células precursoras de oligodendrocitos para volver a un estado multipotente.
  • Para determinar si estas células revertidas pueden auto-renovar y diferenciarse en varios tipos de células neuronales.

Principales métodos:

  • Inducción de células precursoras de oligodendrocitos utilizando señales extracelulares específicas.
  • Evaluación de la capacidad de auto-renovación celular.
  • Análisis de la diferenciación en neuronas, astrocitos y oligodendrocitos.

Principales resultados:

  • Las señales extracelulares pueden inducir a las células precursoras de oligodendrocitos a volver a las células madre neurales multipotenciales.
  • Estas células revertidas exhiben capacidades de auto-renovación.
  • Las células madre neurales multipotentes dan lugar a las neuronas, los astrocitos y los oligodendrocitos.

Conclusiones:

  • Las células precursoras de oligodendrocitos poseen una mayor plasticidad del desarrollo de lo que se reconocía anteriormente.
  • La reversión a un estado multipotente abre nuevas vías para comprender la biología de las células madre neurales y las posibles aplicaciones terapéuticas.
