Sus antecedentes:

  • Las proteínas de señalización requieren una regulación espacial y temporal precisa para efectos celulares localizados.
  • Las pequeñas trifosfatasas de guanosina (GTPSases), como Rac, interactúan con los objetivos aguas abajo y regulan la localización subcelular sobre la unión a GTP.

Objetivo del estudio:

  • Desarrollar un nuevo método para cuantificar la dinámica espacio-temporal de los estados de nucleótidos Rac1 en células vivas.
  • Para investigar el control espacial de la activación Rac inducida por el factor de crecimiento.

Principales métodos:

  • Desarrollo de un indicador de activación de fluorescencia para las proteínas Rho (FLAIR) para el monitoreo en tiempo real.
  • Utilizando FLAIR para visualizar la dinámica del estado del nucleótido Rac1 en las células vivas.

Principales resultados:

  • FLAIR permitió una cuantificación precisa de la dinámica espacio-temporal de Rac1.
  • Control espacial demostrado de la activación Rac inducida por el factor de crecimiento, específicamente en volantes de membrana.
  • Reveló un gradiente de activación de Rac en el borde principal de las células móviles.
