Antigens Involved in Adaptive Immunity

An antigen is any substance the immune system identifies as foreign and potentially harmful to the body, prompting an immune response. Antigens have two functional properties: immunogenicity and reactivity. Immunogenicity is the ability of an antigen to stimulate a specific immune response. At the same time, reactivity describes the antigen's ability to react with the cells and antibodies produced in response to it.

Sus antecedentes:

  • La función precisa del MHC de clase I en el desarrollo neuronal y la plasticidad sigue siendo en gran medida indefinida.

Objetivo del estudio:

  • Investigar el papel de las moléculas MHC de clase I en el refinamiento de las conexiones neuronales durante el desarrollo.
  • Examinar el impacto de la deficiencia de CMH clase I en la plasticidad sináptica en el sistema nervioso central (SNC) de los mamíferos adultos.
  • Para explorar los patrones de expresión de la clase I MHC en distintas poblaciones neuronales.

Principales métodos:

  • Utilizó ratones genéticamente modificados que carecen de superficie celular clase I MHC o el componente CD3zeta de un receptor MHC clase I.
  • Evaluó el refinamiento de las conexiones de la retina a los objetivos centrales durante el desarrollo.
  • Se midió la potenciación a largo plazo dependiente del receptor de N-metil-D-aspartato (LTP) y la depresión a largo plazo (LTD) en el hipocampo de ratones mutantes adultos.
  • Analizó los patrones de expresión de ARN mensajeros MHC de clase I específicos en mosaicos neuronales.

Principales resultados:

  • La deficiencia genética en la clase I MHC o CD3zeta condujo a un refinamiento incompleto de las conexiones entre la retina y los objetivos centrales durante el desarrollo.
  • Los ratones adultos mutantes exhibieron una LTP hipocampal mejorada y una ausencia completa de LTD.
  • Distintos mosaicos de neuronas expresaron ARN mensajeros MHC de clase I específicos, lo que sugiere diversas funciones neuronales.

Conclusiones:

  • Las moléculas MHC de clase I juegan un papel importante en la remodelación dependiente de la actividad de las conexiones neuronales en el desarrollo del SNC.
  • Estas moléculas son críticas para regular la plasticidad sináptica, incluida la potenciación y la depresión a largo plazo, en el SNC maduro.
  • Los hallazgos destacan una función novedosa para el MHC de clase I en el desarrollo y la función del cerebro de los mamíferos.
