Sus antecedentes:

  • El factor de tejido (TF) en monocitos/macrófagos inicia la formación de trombos en los síndromes coronarios agudos.
  • El receptor alfa activado por el proliferador peroxisomático (PPARalpha) regula la expresión génica.
  • Los activadores de PPARalpha pueden reducir la actividad de TF en pacientes.

Objetivo del estudio:

  • Investigar si los activadores de PPARalpha pueden limitar las respuestas de TF en las células monocíticas humanas.
  • Explorar el mecanismo por el cual los activadores de PPARalpha afectan la expresión de TF.

Principales métodos:

  • Los monocitos y macrófagos humanos fueron tratados con activadores de PPARalpha (WY14643, ETYA).
  • El lipopolisacárido (LPS) se utilizó para inducir la actividad y la expresión de TF.
  • Se midió la actividad del TF, la proteína, el ARNm y la actividad del promotor.
  • Se evaluó la unión y la actividad del factor nuclear-kappaB (NF-κB).

Principales resultados:

  • Los activadores de PPARalpha redujeron significativamente la actividad y expresión de TF inducida por LPS en monocitos/macrófagos.
  • Los activadores de PPARgamma no mostraron efectos similares.
  • WY14643 disminuyó la proteína alfa del factor de necrosis tumoral e inhibió la actividad del promotor de TF inducido por LPS, probablemente a través de la inhibición de NF-κB.

Conclusiones:

  • Los activadores de PPARalpha reducen efectivamente la expresión y actividad de TF en monocitos/macrófagos humanos.
  • Estos hallazgos sugieren un papel potencial para los activadores de PPARalpha en la reducción de la trombogenicidad de la lesión aterosclerótica.
  • Los datos ofrecen información sobre cómo los compuestos activadores de PPARalpha pueden afectar la atherothrombosis.
