Sus antecedentes:

  • La supervivencia al injertado de órganos ha mejorado con los últimos fármacos inmunosupresores, pero aún se requiere medicación de por vida.
  • El rechazo del injerto implica la activación de las células T, el reconocimiento de antígenos y la señalización accesoria, siendo la muerte de las células T un mecanismo clave de tolerancia.

Objetivo del estudio:

  • Explorar cómo diferentes fármacos inmunosupresores afectan la tolerancia al trasplante mediado por células T.
  • Diferenciar entre los medicamentos que inhiben la tolerancia al prevenir la muerte de las células T y los que la promueven.

Principales métodos:

  • Análisis de la activación de células T, el reconocimiento de antígenos y las vías de señalización accesorias.
  • Evaluación de los efectos de los fármacos sobre la muerte de las células T (apoptosis) y la falta de respuesta de las células T (anergia).

Principales resultados:

  • Los medicamentos como el micofenolato mofetil, que induce la muerte de las células T, pueden facilitar la tolerancia.
  • La inhibición de la división de células T (por ejemplo, con rapamicina) o la costimulación (por ejemplo, bloqueando CD28 / CD154) puede conducir a la falta de respuesta de las células T y promover la tolerancia.

Conclusiones:
