In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane.

Sus antecedentes:

  • Las infecciones bacterianas gram-positivas representan una amenaza creciente debido al aumento de la resistencia a los antimicrobianos.
  • Los nuevos agentes terapéuticos son esenciales para superar la resistencia en patógenos como Staphylococcus aureus, Enterococcus spp. y Streptococcus pneumoniae.

Objetivo del estudio:

  • Evaluar la eficacia del linezolid, un nuevo agente antimicrobiano de la oxazolidinonona, contra los patógenos gram-positivos resistentes.
  • Evaluar los perfiles farmacocinéticos y toxicológicos del linezolid para su aplicación clínica.

Principales métodos:

  • Pruebas de susceptibilidad in vitro contra patógenos gram-positivos resistentes clave.
  • Análisis de los datos de ensayos clínicos para la neumonía, las infecciones de la piel y los tejidos blandos y las infecciones por enterococos resistentes a la vancomicina.
  • Evaluaciones farmacocinéticas y de seguridad de linezolid.

Principales resultados:

  • El linezolid exhibe actividad bacteriostática in vitro contra el Staphylococcus aureus resistente a la meticilina (MRSA), los enterococos resistentes a la vancomicina (VRE) y el Streptococcus pneumoniae resistente a la penicilina.
  • Eficacia clínica demostrada en el tratamiento de la neumonía, las infecciones de la piel y los tejidos blandos, y las infecciones de ERV.
  • Perfil farmacocinético favorable con una biodisponibilidad oral casi completa y una toxicidad aceptable.

Conclusiones:

  • Linezolid representa un avance significativo en el tratamiento de infecciones graves causadas por bacterias gram-positivas multirresistentes.
  • Ofrece una alternativa viable a las terapias existentes como los glucopéptidos y las estreptograminas.
  • Otras modificaciones de la oxazolidinonona pueden producir agentes con mayor potencia y un espectro de actividad más amplio.
