Resumen

Este estudio demuestra que las interacciones CH ... O ocurren simultáneamente con los enlaces de hidrógeno NH ... O en los derivados de fumaramida, tanto en solución como en estado sólido. Sus antecedentes:

  • Las interacciones no covalentes, particularmente los enlaces de hidrógeno, son cruciales para la estructura y función molecular.
  • Las interacciones CH...O, aunque más débiles que los enlaces N-H...O tradicionales, juegan un papel importante en la estabilización de las conformaciones de péptidos y proteínas.
  • Los derivados de fumaramida sirven como compuestos modelo para investigar estas interacciones.

Objetivo del estudio:

  • Para caracterizar la formación de las interacciones CH...O en los derivados de fumaramida.
  • Para comparar estas interacciones en soluciones y estados sólidos.
  • Establecer derivados de fumaramida como modelos para las interacciones CH...O en estructuras de péptidos y proteínas.

Principales métodos:

  • Análisis de la solución mediante espectroscopia de resonancia magnética nuclear (RMN) de 1H.
  • Análisis de estado sólido mediante cristalografía de rayos X.
  • Experimentos de titulación con dimetilsulfóxido deuterado (DMSO-d6) en cloroformo-d (CDCl3).

Principales resultados:

  • La espectroscopia de RMN de 1H reveló desplazamientos químicos en el campo inferior para los grupos CH involucrados en los enlaces de hidrógeno CH...O=S(CD3) 2, concurrentes con el enlace de hidrógeno NH...O=S(CD3).
  • Los grupos de CH no participantes mostraron cambios mínimos de desplazamiento químico o desplazamientos hacia arriba a concentraciones más altas de DMSO-d6.
  • Las estructuras cristalinas de rayos X confirmaron la presencia de enlaces de hidrógeno CH...O=C junto con enlaces de hidrógeno intermoleculares NH...O=C.

Conclusiones:

  • Tanto la RMN en solución como la cristalografía de estado sólido proporcionan evidencia para el enlace simultáneo de hidrógeno CH...O y NH...O en los derivados de fumaramida.
  • Los enlaces de hidrógeno observados CH...O=C en las estructuras cristalinas se asemejan a los que se encuentran en las estructuras antiparalelas de hojas beta de las proteínas.
  • Los derivados de fumaramida modelan eficazmente las interacciones CH...O relevantes para la estabilidad estructural de los péptidos y las proteínas.
