Sus antecedentes:

  • La enfermedad celíaca es un trastorno autoinmune común que afecta al intestino delgado.
  • La exposición al gluten en individuos genéticamente susceptibles desencadena la enfermedad.
  • El factor de inicio preciso de la respuesta inflamatoria sigue siendo objeto de investigación.

Objetivo del estudio:

  • Para identificar el péptido iniciador primario de la respuesta inflamatoria en la enfermedad celíaca.
  • Para investigar la estabilidad del péptido, la interacción con los autoantígenos y la inmunogenicidad.
  • Explorar estrategias terapéuticas potenciales para la enfermedad celíaca.

Principales métodos:

  • Identificación y caracterización de un péptido específico de 33-mer de gluten.
  • Estudios in vitro e in vivo que evalúan la estabilidad de los péptidos frente a las proteasas digestivas.
  • Los ensayos miden la reactividad del péptido con la transglutaminasa tisular y la inducción de las respuestas de las células T.

Principales resultados:

  • Se identificó un péptido de 33 mer como resistente a la descomposición por las proteasas gástricas, pancreáticas e intestinales.
  • Este péptido reaccionó selectivamente con la transglutaminasa tisular, un autoantígeno clave en el sprue celíaco.
  • El péptido indujo potentemente líneas de células T derivadas del intestino de todos los pacientes con enfermedad celíaca probados y está presente en granos tóxicos.

Conclusiones:

  • El péptido 33-mer identificado es probablemente el principal iniciador de la inflamación inducida por el gluten en la enfermedad celíaca.
  • Su estabilidad e interacción con la transglutaminasa tisular contribuyen a su patogenicidad.
  • La desintoxicación mediante el uso de la bacteriana prolyl endopeptidasa sugiere una posible terapia oral para la enfermedad celíaca.
