Objetivo del estudio:

  • Para sintetizar y caracterizar los miméticos de colágeno ensamblados con una sola cadena, andamio (TRIS) y dendrimero.
  • Para comparar la estabilidad térmica de diferentes secuencias miméticas de colágeno (Gly-Pro-Nleu vs. Gly-Nleu-Pro).
  • Investigar la base estructural para una mayor estabilidad en los miméticos de colágeno ensamblados con dendrímeros.

Principales métodos:

  • Síntesis de péptidos miméticos de colágeno con diferentes secuencias y arquitecturas.
  • Espectroscopia de dicroísmo circular (CD) para evaluar la estructura secundaria y el contenido helicoidal.
  • Estudios de desnaturalización térmica (medidas de la temperatura de fusión) en diferentes disolventes.
  • Estudios de dependencia de la concentración para elucidar los mecanismos de estabilización.

Principales resultados:

  • Los imitadores de colágeno de la secuencia Gly-Nleu-Pro exhibieron una mayor estabilidad térmica que los de la secuencia Gly-Pro-Nleu.
  • Los imitadores de colágeno ensamblados con dendrímeros (162 residuos) mostraron una estabilidad triple helicoidal significativamente mejorada en comparación con las estructuras terminadas en andamios.
  • Se observaron temperaturas de fusión más altas en H2O y 2:1 EG/H2O para los dendrímeros.
  • Los estudios de dependencia de concentración indicaron que el agrupamiento intramolecular estabiliza el haz triple helicoidal al excluir el disolvente.

Conclusiones:

  • La secuencia Gly-Nleu-Pro contribuye a las triple hélices miméticas de colágeno más estables térmicamente.
  • La arquitectura de los dendrímeros mejora la estabilidad de los miméticos de colágeno a través del agrupamiento intramolecular.
  • La exclusión del disolvente por las matrices helicoidales triples agrupadas es el mecanismo principal para la estabilización.
