  • El control de la agregación de tintes dentro de los andamios de ADN es crucial para las aplicaciones funcionales.
  • Los tintes rojo metilo pueden formar agregados H con distintas firmas espectrales.

Objetivo del estudio:

  • Para sintetizar y caracterizar los agregados de metilo rojo H dentro de los conjugados de ADN.
  • Para investigar la influencia de la estructura del ADN en la formación de agregados H y las propiedades espectrales.
  • Para demostrar el control reversible de la estructura del agregado H a través de la hibridación del ADN.

Principales métodos:

  • Síntesis de conjugados de ADN con múltiples fracciones de metil rojo.
  • Espectroscopia UV-Vis para monitorear los cambios espectrales.
  • Estudios de desnaturalización térmica (T(m)) para evaluar la formación de duplex de ADN.

Principales resultados:

  • Se han sintetizado con éxito conjugados de ADN-metil rojo formando agregados H.
  • Se observó una hipsocromicidad significativa y un estrechamiento de la banda con el aumento de la incorporación de colorantes, lo que indica la formación de agregados de H.
  • Cambios espectrales reversibles demostrados (desaparición y reaparición de la banda H) en la formación y disociación del dúplex de ADN.

Conclusiones:

  • Los conjugados ADN-metil rojo pueden formar agregados H bien definidos.
  • Las propiedades espectrales de estos agregados son sensibles a los cambios estructurales del ADN.
  • La hibridación del ADN proporciona un mecanismo reversible para controlar la formación y estructura de agregados de metil rojo H.
