Sus antecedentes:

  • La organogénesis se basa en la morfogénesis de ramificación epitelial, que implica la formación repetitiva de hendiduras y brotes.
  • Los mecanismos moleculares precisos que impulsan estos cambios estructurales, particularmente la formación de hendiduras, siguen siendo en gran medida desconocidos.
  • Las hipótesis anteriores sugirieron la participación de proteínas reguladoras localizadas en la orientación de los patrones de ramificación.

Objetivo del estudio:

  • Para investigar el papel de las proteínas de la matriz extracelular en el inicio de la ramificación epitelial.
  • Determinar la función específica de la fibronectina en la formación de hendiduras durante la morfogénesis ramificada.
  • Para dilucidar los mecanismos moleculares por los cuales la fibronectina influye en la estructura epitelial.

Principales resultados:

  • La expresión de fibronectina fue transitoria y focal en el desarrollo de regiones de hendidura, correlacionándose con una pérdida de cadherina.
  • La supresión de la fibronectina (a través de siRNA o anticuerpos) inhibió la formación de hendiduras y la ramificación en las glándulas salivales, los pulmones y los riñones.
  • La adición de fibronectina exógena promovió la formación de hendiduras y la ramificación.
  • La fibronectina fibrillar indujo adherencias de la matriz celular y pérdida de cadherina en células epiteliales cultivadas.

Conclusiones:

  • La fibronectina es esencial para la formación de hendiduras durante el inicio de la morfogénesis de ramificación epitelial.
  • La fibronectina media una transición crítica de la adhesión célula-célula a la adhesión célula-matriz, impulsando cambios estructurales en los órganos en desarrollo.
  • Estos hallazgos ponen de relieve el papel indispensable de la fibronectina en el desarrollo de arquitecturas complejas de órganos.
