Mechanical Protein Functions

Proteins perform many mechanical functions in a cell. These proteins can be classified into two general categories- proteins that generate mechanical forces and proteins that are subjected to mechanical forces. Proteins providing mechanical support to the structure of the cell, such as keratin, are subjected to mechanical force, whereas proteins involved in cell movement and transport of molecules across cell membranes, such as an ion pump, are examples of generating mechanical force.

  • Las flavin reductasas utilizan residuos de flavina y tirosina donde puede ocurrir la transferencia de electrones fotoinducida.

Objetivo del estudio:

  • Para investigar los cambios estructurales a escala de angstrom en moléculas de proteínas individuales utilizando la transferencia de electrones como una sonda.
  • Para correlacionar las fluctuaciones conformacionales de las proteínas con la reactividad catalítica.

Principales métodos:

  • Utilizó mediciones de fluorescencia de una sola molécula durante toda la vida útil de la flavina.
  • Utilizó la transferencia de electrones inducida por la foto entre la flavina y la tirosina como sonda de distancia.
  • Se realizó un análisis de correlación sobre las variaciones de distancia observadas.

Principales resultados:

  • Variaciones observadas en la distancia flavin-tirosina a lo largo del tiempo a nivel de una sola molécula.
  • Se determinó el potencial de la fuerza media entre la flavina y la tirosina.

  • Las fluctuaciones conformacionales de las proteínas están vinculadas a cambios en la reactividad catalítica.
  • La existencia de múltiples conformadores interconvertidores influye en la función de las enzimas.
