The sense of smell is achieved through the activities of the olfactory system. It starts when an airborne odorant enters the nasal cavity and reaches olfactory epithelium (OE). The OE is protected by a thin layer of mucus, which also serves the purpose of dissolving more complex compounds into simpler chemical odorants. Humans detect odors with the help of specialized cells located in the upper part of the nasal cavity, called olfactory receptor neurons (ORNs). ORNs possess hair-like structures called cilia, which are receptive to sensations from the inhaled air. Principales métodos:

  • Orientación genética en ratones para crear reemplazos e híbridos específicos de la región codificante OR.
  • Análisis de la orientación axonal a los glomérulos en el bulbo olfativo utilizando manipulaciones genéticas M71 y M72 OR.
  • Examen de los polimorfismos de aminoácidos que ocurren naturalmente en las OR y su efecto en la identidad axonal.

Principales resultados:

  • La secuencia de aminoácidos de las OR imparte directamente identidad a los axones OSN, permitiendo su coalescencia en glomérulos.
  • El intercambio de regiones codificantes entre los genes M71 y M72 OR redirigió con éxito los axones a sus glomérulos correspondientes.
  • Las OR híbridas y los polimorfismos naturales revelaron un espectro de fenotipos glomerulares, lo que indica la identidad del axón dependiente del contexto.
  • Los residuos críticos de aminoácidos, principalmente en los dominios transmembrana, son clave para este proceso.

Conclusiones:
