¹H NMR: Complex Splitting

A proton M that is coupled to a proton X results in doublet signals for M. However, NMR-active nuclei can be simultaneously coupled to more than one nonequivalent nucleus. When M is coupled to a second proton A, such as in styrene oxide, each peak in the doublet is split into another doublet.
Splitting diagrams or splitting tree diagrams are routinely used to depict such complex couplings. While drawing splitting diagrams, the splitting with the larger coupling constant is usually applied first.

¹H NMR of Conformationally Flexible Molecules: Variable-Temperature NMR

The axial and equatorial protons in cyclohexane can be distinguished by performing a variable-temperature NMR experiment. In this process, except for one proton, the remaining eleven protons are replaced by deuterium. The deuterium substitution avoids the possible peak splitting caused by the spin-spin coupling between the adjacent protons. The remaining proton flips between the axial and equatorial positions.

The pentose sugar in DNA is deoxyribose, while in RNA the pentose sugar is ribose. The difference between the sugars is the presence of the hydroxyl group on the ribose's second carbon and a hydrogen on the deoxyribose's second carbon.   • Las poliamidas cíclicas se investigan como agentes de unión al ADN.
  • Comprender las interacciones ligando-ADN es crucial para el diseño de fármacos.

Objetivo del estudio:

  • Para determinar la estructura de solución de una poliamida cíclica compleja con un oligómero de ADN específico.
  • Para analizar las interacciones de unión y la dinámica del complejo poliamida-ADN.

Principales métodos:

  • Se utilizó la espectroscopia de Resonancia Magnética Nuclear (RMN) para obtener datos de RMN 2D (1) H.
  • Las restricciones de distancia se derivaron de las intensidades de pico cruzado de NOESY utilizando MARDIGRAS.
  • Se realizaron cálculos de dinámica molecular restringida con un modelo de disolvente para generar estructuras.

Principales resultados:

  • Se determinó la estructura de solución de la poliamida cíclica (ciclo-gamma-ImPyPy-gamma-PyPyPy-) compleja en un duplex de ADN.
  • El ligando exhibe una constante de disociación nanomolar para la unión al ADN.
  • Las estructuras mostraron cierta distorsión del ADN y alteraron el apilamiento de anillos de ligandos debido a la ciclización, pero mantuvieron interacciones de ADN similares en comparación con los complejos no vinculados.
  • Se observó una disociación lenta del ligando y una reducción de las fluctuaciones de apertura del ADN.

Conclusiones:

  • La poliamida cíclica se une de manera estable al ADN, induciendo cambios estructurales menores.
  • La ciclización afecta a la conformación del ligando pero no al perfil general de interacción del ADN.
  • El complejo ligando-ADN demuestra una interacción dinámica entre la unión del ligando y la flexibilidad del ADN.
