Sus antecedentes:

  • El trímero de bobina enrollada N-terminal de gp41 es esencial para la fusión de la membrana viral y celular, un paso clave en la infectividad viral.
  • La inhibición de la interacción entre el trímero gp41 y su compañero de unión C-terminal es una estrategia terapéutica prometedora.
  • El bolsillo hidrofóbico del trimer nativo causa agregación, lo que dificulta su uso como objetivo de cribado.

Objetivo del estudio:

  • Diseñar y caracterizar una imitación eficiente y puramente peptídica del trimer gp41 N-terminal de bobina en espiral.
  • Crear un sistema modelo estable y específico para el estudio de la interacción entre el trímero y el péptido gp41.
  • Desarrollar una plataforma versátil para atacar los mecanismos de fusión viral.

Principales métodos:

  • Diseñado un heterotrimero 1:1:1 utilizando el emparejamiento estérico de las capas del núcleo de alanina/ciclohexilalanina.
  • Incorporado solubilizando pares Glu/Lys y una interfaz específica gp41 en la superficie del trímero.
  • Se utilizaron métodos biofísicos para caracterizar el complejo y su especificidad de unión.

Principales resultados:

  • Se ha creado con éxito una imitación estable y puramente peptídica del trimer de bobina enrollada de bobina gp41 N-terminal.
  • El heterotrimero diseñado se une específicamente al péptido gp41 C-terminal.
  • Un complejo de control que carece de la mitad de la interfaz no mostró ningún vínculo, lo que confirma la especificidad.

Conclusiones:

  • El sistema péptico desarrollado imita efectivamente la función y la especificidad del trímero gp41 nativo.
  • El control de la estequiometría compleja se puede lograr a través de la manipulación de residuos del núcleo interior.
  • Este enfoque ofrece una plataforma versátil para el desarrollo de terapias antivirales y puede extenderse a otros sistemas virales.
