Sus antecedentes:

  • Los complejos de poros nucleares regulan el transporte macromolecular entre el núcleo y el citoplasma.
  • Los importadores y exportadores son los principales transportistas, con RanGTP orquestando la direccionalidad.
  • RanGTP disocia la carga de las importaciones, pero se requiere para la vinculación de exportación en carga.

Objetivo del estudio:

  • Para aclarar las bases estructurales de la exportación de proteínas nucleares.
  • Proporcionar un marco para la comprensión de las distintas funciones de RanGTP en importación y exportación.
  • Para caracterizar el complejo de exportación formado por las exportaciones en Cse1p, Kap60p y RanGTP.

Principales métodos:

  • Cristalografía de rayos X (resolución de 2,0 Å)
  • Análisis estructural del complejo ternario de exportación.
  • Mutagénesis dirigida al sitio.

Principales resultados:

  • La estructura cristalina revela exportin Cse1p en espiral alrededor de RanGTP y Kap60p.
  • Cse1p estabiliza el estado unido a RanGTP y bloquea el dominio de unión a la importina-beta de Kap60p.
  • Mutagenesis muestra cambios conformacionales pareja carga que se une a la afinidad RanGTP, lo que facilita el desmontaje complejo.

Conclusiones:

  • La estructura proporciona un mecanismo molecular para la exportación nuclear mediada por la exportación en Cse1p.
  • Los cambios conformacionales en la exportación son críticos para acoplar la unión de carga y la interacción RanGTP.
  • Este mecanismo garantiza una exportación eficiente y regulada de las importaciones libres de carga.
