  • Para aclarar el papel de los receptores NMDA en la modificación sináptica dependiente de la ubicación.

Principales métodos:

  • Registros electrofisiológicos de la capa cortical de la rata 2/3 neuronas piramidal.
  • Inducción de la plasticidad dependiente del tiempo de picos en diferentes ubicaciones dendríticas.
  • Manipulación farmacológica de los receptores NMDA.
  • Simulaciones por computadora de la plasticidad sináptica y la computación neuronal.

Principales resultados:

  • Tanto la magnitud como la especificidad temporal de la modificación sináptica dependiente del tiempo de picos varían a lo largo de la dendrita apical.
  • Las dendritas distales muestran una potenciación a largo plazo más pequeña y una ventana de tiempo de pico más amplia para la depresión a largo plazo (LTD).
  • La ventana de tiempo de pico para LTD se correlaciona con la supresión del receptor NMDA, que también depende de la ubicación.

Conclusiones:

  • La ubicación dendrítica influye críticamente en la modificación sináptica dependiente del tiempo de picos.
  • La supresión del receptor NMDA es un mecanismo clave que subyace a la inducción de LTD.
  • La inhomogeneidad dendrítica en la plasticidad sináptica mejora la capacidad computacional de las neuronas corticales al permitir la selección diferencial de entradas.
