Sus antecedentes:

  • La modificación permanente del genoma en humanos es un desafío debido a las bajas tasas de recombinación homóloga.
  • Esta limitación obstaculiza la investigación biomédica y el desarrollo de terapias génicas efectivas.

Objetivo del estudio:

  • Desarrollar una solución general para la modificación precisa del genoma in vivo.
  • Para mejorar la frecuencia de recombinación homóloga para aplicaciones de terapia génica.

Principales métodos:

  • Utilizó proteínas de dedo de zinc C2H2 para el reconocimiento de ADN y las diseñó con dominios de nucleasa.
  • Inducción de rupturas de doble hebra del ADN objetivo para estimular la reparación dirigida por homología.
  • Nucleasas de dedo de zinc diseñadas dirigidas a una mutación de inmunodeficiencia combinada severa (SCID) vinculada a X en el gen IL2Rgamma.

Principales resultados:

  • Se logró más del 18% de células humanas modificadas genéticamente sin selección.
  • Se ha demostrado una modificación exitosa en ambos cromosomas X en aproximadamente el 7% de las células.
  • Se observó una reflexión precisa del genotipo a niveles de ARN mensajero y proteínas.
  • Se han reportado altas frecuencias de modificación en células T humanas.

Conclusiones:

  • Las nucleasas de dedo de zinc ofrecen una estrategia viable para la edición precisa del genoma humano.
  • Esta tecnología es prometedora para el desarrollo de nuevas terapias génicas para trastornos genéticos.
  • El enfoque avanza significativamente el potencial para la corrección génica in vivo y el tratamiento de enfermedades.
