Resumen
Las células T CD4+ en reposo resisten la infección por el virus de la inmunodeficiencia humana (VIH) debido a la proteína antiviral APOBEC3G. Esta proteína actúa como un mecanismo de defensa, bloqueando la replicación del VIH-1 y deteriorando la transcripción inversa en células no estimuladas.

Sus antecedentes:
- Las células T CD4+ en reposo son resistentes a la infección por el virus de la inmunodeficiencia humana (VIH), a diferencia de las células activadas.
- El mecanismo detrás de esta resistencia en las células T CD4 + en reposo sigue sin estar claro, con posibilidades que incluyen falta de factores o defensa activa.
Objetivo del estudio:
- Para investigar el papel de APOBEC3G en la protección de las células T CD4 + en reposo de la infección por VIH-1.
- Aclarar el mecanismo por el cual APOBEC3G restringe la replicación del VIH-1 en células T CD4+ no estimuladas.
Principales métodos:
- Análisis de la formación del complejo APOBEC3G (alta masa molecular frente a baja masa molecular) en células T CD4+ en reposo y activadas.
- Tratamiento con RNase para evaluar la actividad APOBEC3G.
- Introducción de ARN pequeños interferentes específicos de APOBEC3G en las células T CD4+ no estimuladas.
- Ensayos de infección por VIH-1 y secuenciación de transcripciones inversas.
Principales resultados:
- APOBEC3G protege fuertemente las células T CD4+ no estimuladas contra la infección por el VIH-1.
- En las células en reposo, APOBEC3G de baja masa molecular (LMM) predomina, bloqueando la replicación del VIH-1 y deteriorando la transcripción inversa.
- La activación conduce al reclutamiento de LMM APOBEC3G en un complejo inactivo de alta masa molecular (HMM), aumentando la permisividad del VIH.
- La reducción de los niveles de APOBEC3G en las células en reposo alivia el bloqueo de replicación temprana para el VIH-1.
- Se observaron bajos niveles de hipermutación de dG dA en las transcripciones inversas, lo que sugiere una restricción independiente de la desaminación.
Conclusiones:
