- Evaluar el potencial de la minociclina como terapéutico para la enfermedad del SNC asociada al VIH.
Principales métodos:
- Los macacos de cola de cerdo infectados con SIV fueron tratados con minociclina o no tratados.
- Se recogieron muestras de sangre y líquido cefalorraquídeo (LCR) y se analizaron la carga viral y los marcadores inflamatorios.
- Se examinó el tejido cerebral en busca de marcadores de encefalitis, inflamación y neurodegeneración.
Principales resultados:
- El tratamiento con minociclina redujo significativamente la gravedad de la encefalitis y los marcadores inflamatorios del SNC.
- Se observó una activación reducida de la proteína quinasa activada por mitógeno p38 y degeneración axonal.
- La minociclina suprimió la replicación viral en el SNC e inhibió la replicación del VIH/SIV in vitro.
Conclusiones:
- La minociclina demostró eficacia en la reducción de la gravedad de la enfermedad del SNC en un modelo de SIV.
- El antibiótico inhibió la carga viral y la neuroinflamación en el cerebro.
- La minociclina es una opción segura, barata y disponible que justifica una mayor investigación como terapéutico anti-VIH.
