Sus antecedentes:

  • La purificación tradicional de proteínas recombinantes a menudo se basa en la cromatografía de afinidad y las proteasas.
  • Estos métodos pueden consumir mucho tiempo, ser costosos y pueden dañar la proteína diana.

Objetivo del estudio:

  • Desarrollar un método simple, rápido y libre de proteasas para la purificación de proteínas recombinantes.
  • Para utilizar un sistema de fusión tripartito con inteína y polipéptido similar a la elastina (ELP) para el aislamiento eficiente de proteínas.

Principales métodos:

  • Una construcción de proteína de fusión que contiene la proteína diana, inteína y ELP se expresó en E. coli.
  • La purificación implicó la agregación inducida por NaCl de la etiqueta ELP, seguida de una microfiltración.
  • La escisión y el aislamiento de la proteína objetivo se lograron a través de la actividad de la inteína y las posteriores transiciones de fase ELP.

Principales resultados:

  • El sistema de fusión tripartita permitió el aislamiento rápido de la proteína diana directamente del lisado de E. coli.
  • El método purificó con éxito las proteínas sin requerir cromatografía de afinidad o proteasas exógenas.
  • Las transiciones de fase reversibles de la etiqueta ELP facilitaron la agregación selectiva y la resolubilización de la proteína de fusión.

Conclusiones:

  • Esta nueva estrategia de purificación ofrece una alternativa simplificada y eficiente para el aislamiento de proteínas recombinantes.
  • El sistema de fusión intein-ELP proporciona una plataforma versátil para la purificación de proteínas, minimizando los pasos de procesamiento y el daño potencial de las proteínas.
