Sus antecedentes:

  • El análisis estructural de la transcripción ha logrado una alta resolución.
  • Las técnicas para manipular moléculas individuales en la transcripción han sido limitadas.

Objetivo del estudio:

  • Desarrollar y aplicar técnicas de manipulación de una sola molécula para estudiar la transcripción.
  • Para dilucidar el mecanismo paso a paso de la ARN polimerasa bacteriana durante la transcripción.

Principales métodos:

  • Se utilizó la espectroscopia de fuerza de una sola molécula para medir el movimiento de moléculas individuales de ARN polimerasa bacteriana.
  • Analizó el tamaño del paso y la dinámica de la translocación de la polimerasa a lo largo del ADN.

Principales resultados:

  • Se demostró que la ARN polimerasa bacteriana se transloca en pasos discretos de un solo par de bases.
  • Se observó que la unión de los nucleósidos trifosfatados entrantes impulsa el movimiento hacia adelante de la polimerasa.

Conclusiones:

  • La ARN polimerasa bacteriana funciona como un ratillo browniano.
  • La energía de unión de los nucleótidos se convierte en trabajo mecánico para impulsar la progresión de la transcripción.
