Sus antecedentes:

  • La comprensión de las propiedades mecánicas del ADN de una sola hebra (ssDNA) es crucial para la biología molecular.
  • No se comprende completamente la influencia del entorno del disolvente en la elasticidad del polímero.
  • El papel del agua en la mediación de las interacciones con biomoléculas como el ADN requiere una mayor investigación.

Objetivo del estudio:

  • Para medir y comparar la elasticidad de una sola cadena de ssDNA en ambientes acuosos y no acuosos.
  • Para aclarar la contribución de los puentes de agua a las diferencias observadas en la elasticidad del ssDNA.
  • Desarrollar un modelo teórico que describa con precisión la elasticidad del ssDNA en diferentes disolventes.

Principales métodos:

  • Se empleó la espectroscopia de fuerza de una sola molécula basada en la microscopía de fuerza atómica (AFM) para medir la elasticidad del ssDNA.
  • Los experimentos se llevaron a cabo en ambientes líquidos acuosos y no acuosos, ambientes líquidos apolares.
  • Se realizaron cálculos de mecánica cuántica ab initio para respaldar los hallazgos experimentales.

Principales resultados:

  • Se observó una desviación marcada en las relaciones fuerza-extensión entre las condiciones acuosas y no acuosas.
  • Esta desviación se atribuyó a la energía requerida para romper los puentes de agua dirigidos por enlaces H alrededor de la cadena de ssDNA en soluciones acuosas.
  • Los resultados en 8 M guanidina-HCl apoyaron aún más el papel de los puentes de agua.
  • Un modelo de cadena de rotación libre sin parámetros coincidía perfectamente con los datos experimentales en disolventes orgánicos.

Conclusiones:

  • Los puentes de agua formados por la unión H entre el ssDNA y las moléculas de agua tienen un impacto significativo en la elasticidad del ssDNA.
  • La ausencia de estos puentes de agua en los disolventes orgánicos conduce a diferentes comportamientos elásticos.
  • El débil enlace H entre el ssDNA y el agua puede ser esencial para la estabilidad del ADN de doble cadena en soluciones acuosas.
