Las arqueas usan la enzima GatDE para amidar el glutamil-tRNA (Gln) en glutaminyl-tRNA (Gln) para la síntesis de proteínas. La estructura cristalina revela un canal de amoníaco y sugiere el reconocimiento de tRNA a través de la forma, no sólo el anticodón, en este vital proceso de código genético.

- Se utilizó la cristalografía de rayos X para determinar la estructura del Methanothermobacter thermautotrophicus GatDE complejado al tRNA (Gln) a una resolución de 3,15 angstroms.
- Se realizaron análisis bioquímicos en mutantes de GatDE y tRNA para caracterizar la actividad enzimática.
Principales resultados:
- La estructura cristalina reveló un canal de 40 angstroms de largo para el transporte de amoníaco que conecta los sitios activos de GatD y GatE.
- El análisis bioquímico identificó los centros catalíticos para las actividades de la glutaminasa, la quinasa y la amidotransferasa.
- tRNA ((Gln) reconocimiento parece estar mediado por la lectura indirecta, que implica la complementariedad de la forma del bucle D.
Conclusiones:
