Los tubos miocárdicos diseñados, creados utilizando tecnología de láminas celulares, exhiben pulsaciones independientes y potencial para el apoyo circulatorio.   • La ingeniería de tejidos ofrece alternativas para el tratamiento de corazones dañados.
  • Los métodos actuales se centran en el trasplante de parches cardíacos.
  • Este estudio explora el uso de hojas de cardiomiocitos para la creación de tubos miocárdicos.

Objetivo del estudio:

  • Para crear tubos miocárdicos pulsables utilizando la ingeniería de hojas celulares.
  • Para evaluar la función in vivo y la supervivencia de estos tejidos modificados.
  • Investigar el potencial para el apoyo circulatorio y la asistencia cardíaca.

Principales métodos:

  • Se utilizaron hojas de cardiomiocitos de ratas neonatales.
  • Se envolvieron sábanas alrededor de la aorta resecada de rata para formar tubos miocárdicos.
  • Se trasplantaron tubos a ratas atímicas para reemplazar la aorta abdominal.

Principales resultados:

  • Los tubos miocárdicos trasplantados mostraron pulsaciones espontáneas y sincrónicas.
  • Se registraron presiones independientes en el injerto de 5.9+/-1.7 mm Hg.
  • La histología confirmó la composición del tejido cardíaco similar a la nativa; se observó mayor espesor y expresión de proteínas específicas en los tubos injertados con aorta en comparación con los injertos de la cavidad abdominal.

Conclusiones:

  • Se pueden diseñar tubos miocárdicos funcionales capaces de apoyar la circulación.
  • La pulsación del flujo sanguíneo del huésped estimula la hipertrofia y el crecimiento de los cardiomiocitos.
  • Esto demuestra un método novedoso para desarrollar tejidos cardíacos diseñados para la asistencia cardíaca independiente.
