Sus antecedentes:

  • Los receptores de inositol 1,4,5-trisfosfato (IP3R) regulan principalmente la liberación de iones de calcio (Ca2+) de las reservas intracelulares.
  • El papel de los IP3R en la mediación de la entrada de Ca2+ a través de la membrana plasmática sigue siendo poco conocido.

Objetivo del estudio:

  • Investigar la función de los IP3R en la entrada de Ca2+, particularmente en respuesta a la activación del receptor de células B (BCR).
  • Determinar la localización y la contribución de las IP3R a la permeabilidad de Ca2+ de la membrana plasmática.

Principales métodos:

  • Electrofisiología de las pinzas de células enteras y de parches perforados en células B de pollo y ratón DT40.
  • Manipulación genética de la expresión IP3R y la función de los poros.
  • Mutagénesis dirigida al sitio y estudios de expresión funcional.

Principales resultados:

  • La activación de IP3R abre un pequeño número de canales Ca2+-permeables en la membrana plasmática (PM).
  • La activación de BCR también evoca la entrada de Ca2+ dependiendo de la función IP3R.
  • Los IP3R se expresan funcionalmente en el PM, contribuyendo significativamente a las señales de Ca2+ evocadas por el BCR.

Conclusiones:

  • Los IP3R están localizados exclusivamente en la membrana plasmática, además del retículo endoplasmático.
  • Una pequeña población de PM IP3R juega un papel crítico en la afluencia de Ca2+ mediada por BCR.
  • Estos hallazgos redefinen el papel de los IP3R en la señalización celular del calcio y las respuestas inmunes.
