Objetivo del estudio:

  • Desarrollar una nueva matriz de lectina fluorescente para la detección de sacarídeos de alto rendimiento.
  • Para utilizar una matriz de hidrogel supramolecular para mejorar la estabilidad y la función de la lectina.

Principales métodos:

  • Construcción de una matriz de lectina fluorescente dentro de un hidrogel supramolecular.
  • Inmovilización no covalente de lectinas fluorescentes bajo condiciones semi-húmedas.
  • Aplicación del apagado y recuperación de fluorescencia bimolecular (BFQR) para la detección de sacarídeos.

Principales resultados:

  • Las lectinas inmovilizadas conservaron capacidades de reconocimiento molecular dentro de la matriz de hidrogel.
  • Lectura fluorescente exitosa de varios sacáridos, incluida la glucosa, los oligosacáridos y las glicoproteínas.
  • Se ha demostrado la detección de alto rendimiento y la elaboración de perfiles de muestras complejas como lisatos celulares.

Conclusiones:

  • La matriz de lectina desarrollada ofrece una plataforma sensible y eficiente para el análisis de glucoconjugados.
  • La matriz de hidrogel preserva eficazmente la función de la lectina, lo que permite una detección robusta de los sacarídeos.
  • Este método simplifica el análisis al eliminar la necesidad de etiquetar el objetivo y los pasos de lavado.
