A decreased body temperature can occur in patients with hypothermia and frostbite. Heat loss with extended cold exposure overpowers the body's ability to create heat, resulting in hypothermia. Sus antecedentes:

  • La deleción clonal en el timo es crucial para la tolerancia de las células T a los autoantígenos.
  • Los mecanismos precisos, la especificidad y los umbrales de afinidad / avidez de la selección negativa siguen siendo incompletamente entendidos.

Objetivo del estudio:

  • Para investigar la especificidad fina y los umbrales de afinidad/avididad de la selección negativa mediada por el receptor de células T (TCR).
  • Para dilucidar la relación entre la fuerza de la interacción TCR y las respuestas de las células T (deleción vs. activación).

Principales métodos:

  • Se utilizaron ratones transgénicos que expresan un TCR específico para el virus de la coriomeningitis linfocítica (LCMV) glicoproteína/H-2Db.
  • Tolerancia inducida al LCMV a través de la infección neonatal con LCMV mutantes que alteran el epitopo de las células T.
  • Se evaluó la deleción de células T en el timo y las respuestas antivirales in vivo e in vitro.

Principales resultados:

  • La inducción de la tolerancia neonatal se logró utilizando LCMVs mutantes.
  • Una interacción TCR-antígeno de baja intensidad fue suficiente para la deleción clonal en el timo.
  • Esta misma interacción de baja avidez no pudo inducir respuestas de células T efectoras en la periferia.

Conclusiones:

  • La selección negativa en el timo opera a un umbral de afinidad/avididad más bajo que la activación de células T.
  • Este umbral diferencial permite la eliminación de células T potencialmente autorreactivas mientras se conservan las respuestas a antígenos extraños.
  • El ajuste fino de las respuestas de las células T se logra a través de distintos requisitos de avidez para la deleción del timo y la activación periférica.
